چکیده :

Background: One of the biggest problems is the increased prevalence of obesity, which is associated with an increased risk of insulin resistance and type 2 diabetes .Visifatin is an adipokine that has recently been isolated from adipose tissue and has insulin-like role in tissues. this study evaluated the effect of Visfatinon Glut4 gene expression in C2C12 mucosal cell lines. Methods: this practical and experimental study Which used C2C12 cells that were capable of differentiating into skeletal muscle to study the viscfatin-activated molecular pathway. The cells tested after replication were divided into four groups, one of which did not receive any material as a control. Three groups were exposed to Visfatin at concentrations of 0.1, 1.0 and 2.0 ng. the expression of Glut4 gene Changes at 24, 48 and 72 hours were evaluated by Real time method. Kidou test, Kruskal-Wallis test, Friedman test were utilized to compare the variables and determine the amount of gene expression changes and statistical analysis of Real Time PCR data by independent and pairwise tests (pared, one-way anova, 2-way anova and Repeated measures ANOVA ). Results: The results showed that expression of Glut4 gene at a concentration of 1.0 ng Visfatin in 72 hours was significantly higher than 0.1 and 2 ng Visfatin concentrations so the high does Visfatin concentration in 72 hours not increase the expression of the Glut4 gene. Discussion: This study showed that visfatin had insulin-like action and activates the expression of Glut4 receptors, which is related to time and Visfatin concentration
