Abstract: Enolase I is expressed in bradyzoite and enolase II in bradyzoite of Toxoplasma gondii. The main
objective of the present study is to identify tachyzoite and bradyzoite forms based on enolase I and II gene
expression using RT-nested PCR-RFLP. Toxoplasma gondii Rh strain tachyzoites were harvested from mouse
peritoneum and bradyzoites (tissue cyst) from brain of rat. Two pair primers were designed for the same
fragments of enolase I and II genes. Total RNA was extracted from both specimens and cDNAs were
synthesized using reverse transcriptase. Nested–PCR was done on cDNAs and PCR products were cut with
TaqI restriction enzyme (RFLP) and the results were run on agarose gel. Identification of tachyzoite and
bradyzoite of Toxoplasma gondii was done successfully using RT-nested PCR-RFLP based on enolase I and
II genes expression.
