چکیده :

Introduction: miRNAs are endogenously small non-coding regulatory RNAs that regulate cognate mRNAs post-transcriptionally. Many miRNAs cloned from mouse stem cells are differentially regulated in various hematopoietic lineages, suggesting that they might influence hematopoietic lineage differentiation. Recent studies have demonstrated that specific miRNAs such as miR-451 have key roles in erythropoiesis. Materials and Methods: Murine embryonic stem cells (mESCs) were infected with lentiviruses containing miR-451. Erythroid differentiation was evaluated using quantitative RT-PCR for hemoglobin chains genes and flowcytometery for erythroid surface antigens. CFU assay was also performed at days 10th , 14th and 21th. Results: MiR-451up-regulation, could induce α, β and γ-globin expression in mESCs and have strong correlation with appearance of CD235a and Ter-119 markers in these cells. Moreover, miR-451 upregulation increases erythroid cells in CFU assay. Conclusions: Our results demonstrate that miR-451 up-regulation strongly induces erythroid differentiation and maturation of mESCs. In the other hands our results provide that expression of miR- 451 have the potential role in erythroid differentiation of mESCs .
