Abstract Rhizospus is a family of opportunistic fungi, and these fungi are separated from many cereals, especially rice and wheat, as well as Asian crops and soil soils. At the end of the Sporangiophores of this Rhizopus Oryzae s, there is a dome-shaped object called Clumella. Around the Clumella is the vesicle, called spongiform encephalitis. Sporangium interferes with a large number of sporulation Spiroglycerides. The samples were cultured on a medium (PDA). For the production of enzymes, an enzyme was used for the solid state fermentation. The effect of carbon content of the initial pH and incubation temperature on the production of enzymes was investigated. Solid state fermentation spore’s suspensions were removed. Enzymes were extracted after fermentation and enzyme activity was evaluated. DNA samples were extracted by boiling method. Designed using a sequence-specific primer. After sequencing, PCR was performed. The genus Rhizopus Oryzae, in most of its species, has high lipase activity and can produce lipase enzymes and, given the wide variety of the presence of this fungus in the country, can be an important genus in biotechnology and industrial activities. Based on the present screening, most native species are capable of producing lipase enzymes and Rhizopus delemar strain BVKT R1 and Rhizopus Oryzae isolate FPZSP368 have high lipase activity and can be promising divisions in the production of this enzyme in a large scale. key words: Enzyme, DNA extraction, lipase, aspergillus, Rhizopus Oryzae
