Background: Reticulocytes are immature red blood cells. A reticulocyte count is a blood test that is used to estimate the degree of effective erythropoiesis, which can be reported as absolute reticulocyte count or as a reticulocyte percentage. Because the reticulocytes do not stain in humid whether, so we decided to solve this problem. Methods: Using a supravital stain (e.g., new methylene blue), residual ribosomal RNA within the reticulocytes is precipitated. An equal volume of stain is added to EDTA-anticoagulated blood, the dilution mixture incubated, a smear is prepared and incubated in incubator 37ºC. The smear is examined to determine the number of reticulocytes present. Results: All of samples that their reticulocytes were not stained by common method, was stained whit our protocol as well as possible. Conclusion: The moisture in the whether obstacle of vital staining and reticulocyte count is not possible.
