The HIV-1 genome encodes 9 genes, comprising gag, pol, env, tat, rev, vif, vpu, vpr and nef. The gag gene encodes a polyprotein precursor whose name, Pr55Gag, is based on its molecular weight. Here, we are describing Gag-polyprotein, starting with its sequence, homology and phylogenetics. Additionally, consisting motifs and structural properties of the Gag-polyprotein were considered. Following their compartmentalization, we studied their genomics, transcriptomics, and proteomics data. We have concluded that CA or p24 peptide of Gag polyprotein has conserved domain that can be utilized for diagnosis of HIV infection. In addition, Gag polyprotein is absolutely essential for packaging the viral particles; if siRNAs or shRNAs could be designed for conserved domains of p24 then virus replication cycle would be interrupted.
